Times Square is near Midtown Manhattan, and Queens is a different borough that is technically on the westernmost part of Long Island. You can take a rather expensive taxi or shuttle between the two (might take half an hour to an hour, depending on the time of day/rush hour), or the train from Times Square to whatever part of Queens you are trying to visit. You might take the E or F trains, then a bus/taxi/Uber/walk...it depends. Are you going to a specific neighborhood in Queens? The airport?

So Katie, settling in Queens to meet NYC isn't practical. It is in the hands of the tourist centres.
You could stay in a hotel or Airbnb in Long Island City (technically part of Queens), which has quick access to many parts of Manhattan by Uber/Lyft/taxi, the train, and possibly even biking. Be very careful if you bike in Manhattan. Car drivers will be aggressive.
